MTAP, a crucial enzyme, facilitates the salvage pathway for adenine and methionine synthesis. It breaks down 5'-methylthioadenosine into adenine and 5-methylthio-D-ribose-1-phosphate, which are then converted into AMP and methionine, respectively (1,2). Present in all normal cells and tissues, MTAP is often lost in various human tumors like pancreatic adenocarcinoma, neuroendocrine tumors, non-small cell lung carcinoma, and breast carcinoma. It's usually codeleted with p16 (cdkN2a/ARF) (3-5). Overexpression of MTAP in breast cancer cells inhibits colony formation in soft agar, suggesting its role as a tumor suppressor (6).
